ABSTRACT

ABSTRACT: This study evaluated the behavior and welfare conditions of laying chicks in the starter phase, fed with diets formulated with increasing levels of crude fiber (CF) and two feedstuffs with different fiber compositions. The experiment was developed at the Federal University of Goiás, in Goiânia - GO, Brazil. Three hundred Bovans White chicks were evaluated in a completely randomized design with a 2×2+1 factorial arrangement corresponding to two levels of CF in the diet (3.0 and 3.5%) and two fiber sources (wheat bran and sugarcane bagasse), plus a control treatment (maize- and soybean meal-based), totaling five treatments with six replicates of 10 birds each. Maintenance behavioral patterns and welfare were assessed at 21 and 42 days of age. There was no difference in behavior and classification by order of importance of the activities performed by the fiber-fed birds observed at 21 and 42 days, according to the Kruskal Wallis test (P>0.05). Medium and good welfare conditions were obtained with wheat bran at 3.5 and 3.0% CF, respectively, at 21 days of age. At 42 days, the use of sugarcane bagasse at the CF level of 3.5% and wheat bran at the level of 3.0%provided good behavior conditions. The use of crude fiber in the feeding of hens did not change their behavior.


RESUMO: Objetivou-se avaliar o comportamento de pintainhas de postura na fase de cria, alimentadas com dietas formuladas com aumento de fibra bruta e dois alimentos com composições de fibra diferentes. O trabalho foi realizado na Universidade Federal de Goiás, Goiânia, GO, utilizando-se 300 pintainhas Bovans White, em delineamento inteiramente casualizado e arranjo fatorial 2x2+1, com dois níveis de fibra bruta na dieta (3,0% e 3,5 % de FB), duas fontes de fibra (farelo de trigo e bagaço de cana) e o tratamento testemunha (ração a base de milho e farelo de soja), totalizando cinco tratamentos e seis repetições de 10 aves cada. Foram avaliados padrões de comportamento de manutenção e bem-estar aos 21 e 42 dias de idade. Não houve diferença no comportamento e na classificação por ordem de importância das atividades realizadas pelas aves alimentadas com fibra e observadas aos 21 e 42 dias pelo teste de Kruskal Wallis (P>0,05). Obteve-se condições média e boa com farelo de trigo e 3,5 e 3,0% de fibra bruta, respectivamente, com 21 dias de idade. Aos 42 dias, o uso de bagaço de cana ao nível de 3,5% de fibra bruta e o farelo de trigo ao nível de 3,0%, mostraram boas condições de comportamento. A utilização de fibra bruta na alimentação de frangas não alterou o comportamento das aves.

ABSTRACT

This investigation was conducted to evaluate the effect of incorporation of wheat bran (WB) with dried apple pomace (DAP) or dried carrot pomace (DCP) on quality attributes of chicken nuggets. Raw emulsion was stuffed in rectangular moulds, cooked in electric oven at 165°C for 40 minutes and nuggets were prepared by slicing cooked emulsion to 2-3 cm3 size. Colour scores increased with increase in DAP level in WB + DAP treated nuggets. A steady decrease in scores for all other sensory attributes was noticed in both WB + DAP and WB + DCP treated nuggets with increase in fibre level. Hardness and shear press value increased whereas springiness and cohesiveness decreased with increase in fibre level. Gumminess and chewiness of WB + DAP treated nuggets increased with increase in fibre level while gumminess and chewiness of WB + DAP treated nuggets showed a variable trend. Cooking yield and emulsion stability of fibre enriched nuggets were significantly higher whereas moisture, protein, cholesterol content and pH were significantly lower than control. Insoluble, soluble and total dietary fibre content increased drastically in treated nuggets. Thiobarbituric acid reacting substance (TBARS) value and microbial counts increased significantly during refrigerated storage in all the products. It is concluded that healthier and dietary fibre enriched chicken nuggets with refrigerated storage stability up to 15 days can be prepared using combination of wheat bran (2%) with dried apple pomace (6%) or dried carrot pomace (4%) each

ABSTRACT

Abstract This study evaluated the production of endoxylanases by Streptomyces malaysiensis AMT-3 in submerged fermentation using by-products of the food industry at 28ºC. In shake-flasks experiments, the highest endoxylanase activity of 45.8 U.mL-1 was observed within 6 days in a medium containing (w/v) 2.5% wheat bran and 1.2% corn steep liquor. The same culture conditions were used to evaluate the enzyme production in a 2 L stirred tank reactor under different agitation (300, 450 and 600 rev.min-1) and aeration (30 and 60 L.h-1) conditions. The use of 450 rev.min-1 coupled to an aeration of 90 L.h-1 resulted on 81.3 U.mL-1 endoxylanase activity within 5 days. The effect of temperature and pH on endoxylanase activity and stability showed the highest activity at 60 ºC and pH 6.0. Zymography showed the presence of three xylanolytic bands with molecular masses of 690, 180 and 142 kDa. The results showed that the thermotolerant actinobacterial endoxylanase can be produced in high titers using by-product of the food industry.

ABSTRACT

Objective:To quickly analyze and identify the differential chemical compositions of Aurantii Fructus Immaturus before and after stir-frying with bran and chemical compositions of wheat bran after processing by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MSE) combined with UNIFI database screening method. Method:ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 µm) was used for chromatographic separation with 0.1% formic acid solution (A)-acetonitrile (B) as the mobile phase for gradient elution (0-11 min, 98%-70%B; 11-15 min, 70%-55%B; 15-16 min, 55%-35%B; 16-20 min, 35%-5%B; 20-20.5 min, 5%-98%B; 20.5-22 min, 98%B) at the flow rate of 0.3 mL·min-1 and the injection volume of 2 µL. The analytes were determined in positive ion mode with electrospray ionization (ESI) and data collection range of m/z 50-1 500. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were used to find the component differences between raw and processed products of Aurantii Fructus Immaturus, and the chemical compositions of wheat bran after processing were determined. Result:There were 64 compounds in raw products, 58 compounds in bran-fried products, and 18 compounds in wheat bran.There were 19 different components between raw and processed products of Aurantii Fructus Immaturus, mainly volatile oil, flavonoids, phenolic acid, coumarins and saponins. Conclusion:Based on the analysis of these different components before and after stir-frying with bran and the chemical compositions carried by wheat bran, the stir-frying with bran can alleviate the intensity of Aurantii Fructus Immaturus, which proves the necessity of stir-frying with bran for the processing technology of this herb, and provides a comprehensive experimental basis for research on processing mechanism of Aurantii Fructus Immaturus.

ABSTRACT

ABSTRACT: The aim of this study was to measure the nitrogen (N) balance of pigs fed with lysine-limiting diets containing practical levels of wheat bran (WB) or soybean hulls (SH). Twelve pigs with average weights of 57.36±2.01 and 72.68±3.24 kg were used in trials 1 and 2, respectively. In trial 1, treatments were CT1 - control diet and WB - diet with inclusion of 15% WB. In trial 2, CT2 - control diet and SH - diet with inclusion of 6% SH. Fibrous diets increased (P<0.05) the fecal N in 63.54 and 60.55% in relation to CT1 and CT2, respectively. The urinary N was higher (P<0.05) in pigs receiving the WB diet, but was not influenced in the trial with SH. The N retention (NRET) was higher (P>0.05) in pigs ingesting the WB diet; although, when the proportion of NRET was expressed relative to N ingested (NING) there was no difference (P>0.05) between treatments. Inclusion of SH did not affect (P>0.05) the NRET when the result was expressed in absolute or proportional terms. In conclusion, inclusion of practical levels of WB and SH does not significantly affect the metabolic costs involved with nitrogen metabolism. However, only WB was able to contribute to the N balance of pigs, while the relative amount of N added by SH was fully recovered in feces.


RESUMO: O objetivo deste estudo foi medir o balanço de nitrogênio (N) de suínos alimentados com dietas limitantes em lisina contendo níveis práticos de inclusão de farelo de trigo (FT) ou casca de soja (CS). Foram utilizados 24 suínos machos castrados com peso médio inicial de 57,36±2,01 kg e 72,68±3,24 kg nos experimentos 1 e 2, respectivamente. No experimento 1 os tratamentos foram CT1 - dieta controle, e FT - dieta com inclusão de 15% de FT. No experimento 2 CT2 - dieta controle, e CS - dieta com inclusão de 6% CS. As dietas fibrosas aumentaram (P<0,05) o N fecal em 64,54 e 60,55% em relação a CT1 e CT2, respectivamente. O N urinário foi maior (P<0,05) nos suínos que receberam a dieta FT, mas não foi afetado no experimento com CS. A retenção de N (NRET) foi maior (P<0,05) nos suínos alimentados com a dieta FT, entretanto quando a proporção do NRET foi expressa em relação ao N ingerido (NING) não houve diferença entre os tratamentos (P<0,05). A inclusão de CS não afetou (P<0,05) a NRET quando os resultados foram expressos em termos absolutos ou proporcionais. Em conclusão, a inclusão de níveis práticos de FT ou CS não afeta significativamente o custo metabólico envolvido no metabolismo de N. Entretanto, apenas o FT foi capaz de contribuir para o balanço de N dos suínos, enquanto a quantidade N relativa a adição de CS foi totalmente recuperada nas fezes.

ABSTRACT

The present study deals with the isolation screening and optimization of fungal strain for pectinase production. The fungal strains were isolated from different sources, including soil, fruits etc. Qualitative screening was performed on the basis of the pectin hydrolysis zone. While, quantitative screening was carried out employing submerged fermentation. Among all the strains the strains showing highest pectinolytic potential were selected identified and assigned the code Aspergillus niger ABT-5.The influence of different fermentation media on pectinase production was evaluated. The M5 medium containing 10g wheat bran, nutrient medium containing (g/l) of (NH4)2SO4 6.0, K2HPO4 6.0, KH2PO4 6.0, MgSO4.7H2O 0.1 gave the highest pectinase production. The other important physico chemical parameters including incubation period, temperature, and volume of media, size of inoculum, carbon and nitrogen sources were also optimized for pectinase production. The highest pectinase production (15.5U/ml) was obtained at 72h of incubation, pH 6, temperature 30°C, volume of media 50ml. Fructose and urea were designated as best carbon and nitrogen sources subsequently.


O presente estudo trata da triagem de isolamento e otimização da cepa fúngica para produção de pectinase. As cepas fúngicas foram isoladas de diferentes fontes, incluindo solo, frutas, etc. A triagem qualitativa foi realizada com base na zona de hidrólise da pectina. Enquanto, a triagem quantitativa foi realizada utilizando fermentação submersa. Entre todas as cepas, as cepas que apresentaram maior potencial pectinolítico foram selecionadas e atribuídas ao código Aspergillus niger ABT-5. Avaliou-se a influência de diferentes meios de fermentação na produção de pectinase. O meio M5 contendo 10g de farelo de trigo, meio nutriente contendo (g / l) de (NH4)2SO4 6.0, K2HPO4 6.0, KH2PO4 6.0, MgSO4.7H2O 0.1, proporcionou a maior produção de pectinase. Os outros parâmetros físico-químicos importantes, incluindo período de incubação, temperatura e volume dos meios, tamanho do inóculo, fontes de carbono e nitrogênio também foram otimizados para a produção de pectinase. A maior produção de pectinase (15,5U / ml) foi obtida às 72h de incubação, pH 6, temperatura 30 ºC, volume dos meios 50ml. A frutose e a ureia foram designadas como melhores fontes de carbono e nitrogênio posteriormente.

ABSTRACT

Aim: The study of laccase production by Trametes pubescens cultured on cladode extractive residues alone, and as co-carbon source with wheat, using submerged fermentation condition. Place and Duration of Study: Biotechnology Laboratory, Durban University of Technology, Durban, South Africa. Methodology: Plant extraction, submerged fermentation, enzyme activity and characterization techniques were utilized. Results: The highest laccase activity was observed at 60-80% wheat: residue ratio combinations under submerged conditions with copper and xylidine supplementation. Partially purified enzyme fractions showed similar characteristics at different temperatures and pHs. There was good retention of relative enzyme activity at temperatures near 60°C, and pH stability from pH 4.0-6.0. At optimal culture conditions laccase activity was highest at 49.01±0.21 U/ml (80% wheat: 20% residue ratio), and lower at 10.02±0.51 U/ml (80% wheat:20% NEP residue). The optimum temperature for laccase fractions was 25°C and pH optimum at 5. Highest specific activity was 3.55 U/mg of protein for the 80% wheat: 20% residue ratio laccase extract. Conclusion: These results show the potential of cladode extractable phenol residues as potential economic growth medium for laccase production, offering a new alternative use for this agro-industrial and/or laboratory by-product.

Objective: To explore the effect of different processing methods on the content changes of ferulic acid and total flavonoids in wheat bran and its processed products by HPLC and UV spectrophotometry.Methods: Ferulic acid was determined by HPLC with SHIMADZU C18 (250×4.6 mm,5 μm) as the analytical column, the mobile phase was composed of acetonitrile-0.1% phosphoric acid solution (16∶84), the flow rate was 1.0 ml·min-1, the detection wavelength was 320 nm and the column temperature was 35℃.Total flavonoids was determined by UV spectrophotometry at the wavelength of 258 nm(±2 nm).Results: The content of ferulic acid in the crude bran, the toasted to brown bran, the toasted to dark brown bran, the over toasted bran and the honey toasted bran was 2.07, 2.33, 1.68, 1.11 and 0.83 μg·g-1, and total flavonoids was 5.96, 7.15, 9.73, 13.61 and 9.42 mg·g-1, respectively.Conclusion: The higher the toasted processing degree is, the lower content of ferulic acid and the higher content of total flavonoids are in wheat bran.

Objective:To research the best processing method for Bupleurum chinense DC.by orthogonal tests.Methods:With the contents of saikosaponin a and saikosaponin d as the indices,the L9(34) orthogonal table was used to study three factors including the amount of wheat bran,pot temperature before heating and processing time.The orthogonal design was applied to study the processing technology of Bupleurum chinense DC.fried with wheat bran.Results:The best processing method was as follows:100 g Bupleurum chinense DC.was mixed with 10 g wheat bran and fried at 290 ℃ for 80 seconds.Conclusion:The optimized processing technology is reasonable,reliable and highly reproducible,which provide reference for the processing of Bupleurum chinense DC.with wheat bran.

ABSTRACT

Beauveria bassiana is an entomopathogenic fungus with high potential in controlling insect pests. In this study, we propose optimum cultural conditions and culture media for better growth of various B. bassiana strains. B. bassiana strains achieved their maximum growth during optimal incubation period of seven days. The optimum pH and temperature for maximal growth of B. bassiana strains was found to be 6-7 and 25-30ºC, respectively. All the tested carbon and nitrogen sources supported growth and development of the B. bassiana strains. Starch and peptone as carbon and nitrogen sources supported maximum radial growth (2.13-3.00 cm) and conidiospore count in both solid state culture (2.66×107 conidia/mL) and liquid state culture (9.86×107 conidia/mL). Strain BbR2 was the fastest growing strain on almost all nutrient sources studied and possessed commendable growth rate and sporulation potential. Wheat bran (WB) and rice bran (RB) in the proportion of 3:1 supported maximum conidiospores yields (1.90×107 conidia/mL) for strain BbR2 in solid state fermentation conditions.

ABSTRACT: In order to evaluate the efficiency of phytase in diets with low and high phytate phosphorus (PP) 0content, as a consequence of wheat bran inclusion, on the relative weight of organs, intestinal morphometry and performance, three hundred and eighty-four male Cobb500 broilers were housed in metabolic cages. Animals were assigned into four treatments in a 2x2 factorial scheme in a randomized block design with eight replicates of 12 birds each. From 11 days of age birds received experimental diets, which consisted of: Diet low in PP; Diet low in PP with phytase (500FTU kg-1); Diet with a high PP and Diet with a high PP with phytase (500FTU kg-1). At 22 and 32 days of age two birds were slaughtered in order to collect gizzard, heart, liver, cecum, cloacal bursa, and at 32 days, a portion of the duodenum, jejunum and ileum was collected for morphometric evaluation. From 22 to 32 days of age average feed intake, average weight gain, average body weight and feed conversion ratio were also evaluated. Data were subjected to analysis of variance, fixed effects of diet and phytase and interaction between factors as well as the random block effects were tested. There was no significant interaction for the variables studied, concluding that phytase in diets with low or high phytate phosphorus content did not change the relative weight of organs, intestinal morphometrics and performance; only isolated effects were observed.


RESUMO: Para avaliar a eficiência da fitase em dietas com baixo e alto teor de fósforo fítico (PP), em função da inclusão ou não do farelo de trigo, sobre o peso relativo de órgãos, morfometria intestinal e desempenho, foram alojados 384 frangos de corte, machos da linhagem Cobb500, em gaiolas metabólicas. Os animais foram distribuídos em quatro tratamentos em um arranjo fatorial 2x2 em delineamento de blocos casualizados com oito repetições e 12 aves por unidade experimental (UE). A partir de 11 dias de idade as aves receberam as dietas experimentais, que consistiram em: Dieta com baixo teor de PP; Dieta com baixo teor de PP com fitase (500FTU kg-1); Dieta com alto teor de PP e Dieta com alto teor de PP com fitase (500FTU kg-1). Aos 22 e 32 dias de idade foram abatidas duas aves por UE para coletar a moela, coração, fígado, ceco, bolsa cloacal, e aos 32 dias foi coletada uma porção do duodeno, jejuno e íleo para avaliação da morfometria. No período de 22 a 32 dias de idade foram avaliados o consumo médio de ração, ganho de peso médio, peso médio corporal e a conversão alimentar. Os dados foram submetidos à análise de variância, onde foram testados os efeitos fixos de dieta e fitase e a interação entre os fatores, bem como o efeito aleatório de bloco. Não foi observada interação significativa para nenhuma das variáveis estudadas, concluindo-se que a fitase em dietas com baixo ou alto de PP não altera o peso relativo dos órgãos, a morfometria intestinal e o desempenho, apenas efeitos isolados foram observados.

ABSTRACT

Twenty four fungal species were screened for their ability to produce alkaline L-methioninase on methionine-glucose liquid medium. Aspergillus ustus AUMC 10151 displayed the highest yield of enzyme (10.8 U/mg protein), followed by A. ochraceus and Fusarium proliferatum. Upon optimization of the submerged fermentation (SmF)conditions, the maximum enzyme yield (18.23 U/mg protein) was obtained on a medium containing L-methionine (0.5%), sucrose (0.95%), KH2PO4 (0.1%) and 175 rpm. Seven agro-industrial by-products were screened as substrates for L-methioninase production under solid-state fermentation (SSF). Wheat bran resulted 38.1 U/mg protein, followed by rice bran (27.6 U/mg protein) and soya bean meal (26.6 U/mg protein). Maximum alkaline L-methioninase (99.56U/mg protein) was achieved at initial moisture content of 71.5%, inoculum size of 2.0 mL of spore suspension, initial pH 8.5, incubation period eight days at 30°C and supplementation of the salt basal medium with pyridoxine(100 μg/mL) and beet molasses (20% v/v). The productivity of L-methioninase by A. ustus under SSF was higher than that of SmF about 5.45 fold under optimum conditions.

ABSTRACT

The aim of this study was to evaluate peptidase production by Aspergillus terreus in solid-state bioprocess and evaluate its parameters. The best conditions were 5.0 g of wheat bran as substrate, incubation temperature 30°C, inoculum 2.0x105 spores/g and 75% saline volume, with production reaching 677 U/mL (5400 U/g culture medium) after 72 h of fermentation. Biochemical characterization of the crude enzymatic extract showed the optimum pH and temperature of 6.5 and 55°C, respectively. The stability at different temperatures and pH values showed that the extract could endure different pH. The evaluation of the ions influence and inhibitors proved that the enzyme required an ion for better activity, which was corroborated with the inhibition of EDTA and PMSF, characterizing serine and/or metallo peptidase. The extract was also tested for specific activities and showed promising results for keratinolytic and collagenolytic activities (0.252 and 0.165 OD/mL, respectively).

ABSTRACT

Studies were conducted on the production of protease by moderately halophilic Bacillus sp. on agro-industrial waste materials. The bacterium could efficiently use many agro wastes as substrates but wheat bran supported maximum enzyme production. To ascertain the performance of the process in shake flasks and lab scale bioreactor, experiments were conducted to analyse protease activity utilizing wheat bran as cost effective substrate. The studies unveiled that pH 7.0, temperature 30°C and static conditions were optimal for enzyme production in flask level fermentation. In scale-up fermentation, at optimal pH and temperature, agitation rate of 50 rpm was best for protease production. The enzymatic nature was studied in 10% SDS gels with BSA (2.5 mg/mL) as substrate and banding pattern was compared with undigested BSA as control. The endoprotease nature and the kinetics of protease activity were confirmed. The enzyme retained 37% of its activity even at 5 M NaCl concentration. The proteolytic activity was also confirmed by casein zymogram analysis. The fermentation medium containing inexpensive substrates, physical conditions and ability of Bacillus sp. to exhibit protease activity on a large scale could collectively be useful for commercial production.

Objective To optimize the processing technology for Radix Puerariae simmered with wheat bran.Methods Orthogonal experiment L9(34) was chosen to optimize the technology. The external properties of Radix Puerariae simmered by wheat bran, the content of puerarin and the antidiarrheal effect on mice with diarrhea caused by folium sennae were used as indexes. Comprehensive weighted score was employed to optimize simmering Radix Puerariae with wheat bran technology.Results Processing time was the main affecting factor, while processing temperature had no significant effect. The optimum processing parameters were 100 g Radix Puerariae simmered with 30 g wheat bran at 160℃ for 2 minutes.Conclusion The optimum processing technology was simple and convenient, and with good reproducibility and operability. It is also helpful for the quality control of Radix Puerariae simmered with wheat bran.

ABSTRACT

The influence of various oil cakes has been investigated for high level production of lipase using Aspergillus tamarii MTCC 5152. By solid state fermentation in wheat bran containing 2.5% w/w gingili oil cake at 70% v/w moisture content the fungus produced a maximal yield of lipase (758 ± 3.61 u/g) after 5 days of incubation using 2% v/w inoculum containing 10(6) spores/mL. Wheat bran and gingili oil cake with supplementation of gingili oil (1.0% w/w), glucose (0.5% w/w) and peptone (0.5% w/w) gives an increased enzyme production of 793 ± 6.56 u/g. The enzyme shows maximum activity at pH 7.0, temperature 50 °C and was stable between the pH 5.0-8.0 and temperature up to 60 °C. Crude lipase (3%) applied to tannery fleshing shows 92% fat solubility. The results demonstrate that fat obtained from tannery fleshing, a by-product of the leather industry has a high potential for biodiesel production and the proteinaceous residue obtained can be used as animal feed.

ABSTRACT

The present study reveals that a potent laccases producer Pycnoporus cinnabarinus produces higher quantity of laccases in the Coll et al. Medium (M1 medium) and Potato dextrose broth medium (M4 medium) containing agricultural waste ‘wheat bran’ (1%w/v) after a great comparative study among the five laccases production media namely Coll et al. medium (M1 medium), Olga et al. medium (M2 medium), Slomczynski medium (M3 medium), Potato dextrose broth (PDB) medium (M4 medium) and nutrient salt medium (M5 medium). There was a tremendous increase in laccases production with alternative carbon source. There was about 265.2% increase in laccases production in M1 Medium with wheat bran (126.99±2.046 U/ml/min) which further increased by 305% (140.78±1.118 U/ml/min) after the addition of dairy waste ‘Ghee residue’. In the presence of rice bran there was a reduction in laccase production, which however improved in the presence of Ghee residue by a good 63.8% (56.96±1.005 U/ml/min) compared to the M1 medium with glucose. Similarly in case of M4 medium, there was an increase in laccases production by 45.6% with wheat bran (89.46±1.861U/ml/min) and on addition of ghee residue it increased to about 95.6% (115.00±2.048U/ml/min). The beneficial effect of ghee residue was proved by comparative study on effect of other inducers namely copper sulfate, ammonium tartrate, ethanol and peptone. The maximum production was obtained in 9 days at pH 3.0 and temperature 35°C in potato dextrose broth medium (M4) with 1% w/v wheat bran and 2% w/v Ghee residue. The presence of phenolic compounds in the Ghee residue was found to be encouraging laccases production.

ABSTRACT

Aim: The objective of the present work was to optimize the environmental parameters for Cellulase (1,4 β-endoglucanase, E.C.3.2.1:4) production using Penicillium citrinum MTCC 9620 in Solid State Fermentation. Study Design: One unit of Cellulase (1,4 β-endoglucanase, E.C.3.2.1:4) activity is defined as the amount of enzyme producing 1μmole of glucose equivalent/min measured using UV visible spectrophotometer at 540 nm. Place and Duration of Study: Food Technology laboratory of Dr. S. S. Bhatnagar University Institute of Chemical Engineering & Technology, Panjab University, Chandigarh between January and June 2011. Methodology: Penicillium citrinum MTCC 9620 was maintained on potato dextrose agar (PDA) at 4°C. For Cellulase production Czapek Yeast Extract medium was used as moistening medium. Incubation temperature, pH, incubation time and other parameters like suitable substrate, pre-treatment of the substrate on production of Cellulase in Solid State Fermentation (SSF) was optimized using agricultural residues by Penicillium citrinum MTCC 9620. Microscopic and Spectral properties of substrates were determined to detect the structural changes after pre-treatment. Results: Production of extracellular Cellulase was greatly affected by variation in substrates, pre-treatments of substrate and variation in pH and incubation temperature. Cellulase activity was significantly (p < 0.05) higher when alkali treated wheat bran was used as substrate than untreated substrate. Among three substrates and their three pretreatment conditions, It has been observed that alkali treated wheat bran was the most suitable substrate for maximum cellulase production (12.56± 0.097U/mL) at pH 5.5 and 30ºC without any extraneous nitrogen source by Penicillium citrinum MTCC 9620 after 120 h of fermentation time. SEM study revealed that during alkali treatment the solid surface become rough which results growth of fungus eventually maximum cellulase production. Conclusion: P. citrinum MTCC 9620 is one of the potential cellulase producing fungal strain. Optimum condition of cellulase (1,4 β-endoglucanase, E.C.3.2.1:4) production by P. citrinum MTCC 9620 was 30°C temperature, 5.5 pH when alkali treated wheat bran was used as substrate. Growth kinetics of P. citrinum MTCC 9620 was studied and it showed adequacy of fit to Monod Model to describe the growth pattern of P. citrinum MTCC 9620 in SSF at 30°C for 120 h incubation period.

ABSTRACT

This work sought to evaluate the ability of fungi Phanerochaete chryosporium to degrade congo red azo dye in batch reactor, evaluate the influence of glucose and wheat bran as co-substrates on the removal of the dye in the medium and investigate the influence of ammonium chloride, ammonium nitrate and ammonium sulfate as the inorganic nitrogen source for the process. Wheat bran was not effective satisfactorily for the removal of dye and organic matter had no desired effect for the removal of color and organic matter and showed the lowest values of k2, 0.008 M-1.d-1and 0.0004 M-1.d-1, respectively. Glucose presented the best response with the highest final percentage of dye removal (97%) and rate of dye removal (0.017 M-1.d-1), without adding an external source of nitrogen.

ABSTRACT

Polyhydroxyalkanoates (PHA) and α-amylase (α-1,4 glucan-4-glucanohydrolase, E.C. 3.2.1.1) were co-produced by Bacillus sp. CFR-67 using unhydrolysed corn starch as a substrate. Bacterial growth and polymer production were enhanced with the supplementation of hydrolysates of wheat bran (WBH) or rice bran (RBH) individually or in combination (5-20 g L-1, based on weight of soluble substrates-SS). In batch cultivation, a mixture of WBH and RBH (1:1, 10 g L-1 of SS) along with ammonium acetate (1.75 g L-1) and corn starch (30 g L-1) produced maximum quantity of biomass (10 g L-1) and PHA (5.9 g L-1). The polymer thus produced was a copolymer of polyhydroxybutyrate-co-hydroxyvalerate of 95:5 to 90:10 mol%. Presence of WBH and corn starch (10-50 g L-1) in the medium enhanced fermentative yield of α-amylase (2-40 U mL-1 min-1). The enzyme was active in a wide range of pH (4-9) and temperature (40-60ºC). This is the first report on simultaneous production of copolymer of bacterial PHA and α-amylase from unhydrolysed corn starch and agro-industrial residues as substrates.
